#1 - Enzootic calcinosis in sheep in Uruguay: a brief review and report of two outbreaks

Abstract in English:

This study aimed to do a brief review of enzootic calcinosis in sheep and to report two outbreaks of Nierembergia rivularis poisoning in sheep in Uruguay. The outbreaks occurred in farms located on an island (Outbreak A) and on the border (Outbreak B) of the Rincón del Bonete lake. Sheep of all ages were affected, with the exception of suckling lambs. The first clinical signs occurred in early October, and deaths occurred from December to February. Outbreaks A and B had morbidity of 10%, and the mortality was 7.2% and 2.8% in Outbreaks A and B, respectively. The clinical signs included weight loss, retracted abdomen, stiff gait, and kyphosis. An autopsy was performed on one sheep from each outbreak. Pulmonary and arterial calcification, nephrocalcinosis, and osteopetrosis were observed in gross and microscopic examination in both sheep. Thyroid C-cell hyperplasia and carcinoma was observed in sheep A. Sheep B showed thyroid C-cell hyperplasia and parathyroid chief cell atrophy. The parathyroid was not examined in the sheep from Outbreak A. The differential diagnosis of enzootic calcinosis in southern South America should consider four toxic plants in the Solanaceae family: Solanum glaucophyllum, Solanum stuckertii, Nierembergia veitchii, and Nierembergia rivularis.

Este trabalho faz uma breve revisão da calcinose enzoótica em ovinos e descreve dois surtos de intoxicação por Nierembergia rivularis em ovinos no Uruguai. Os surtos ocorreram em propriedades localizadas em uma ilha (Surto A), e nas margens (Surto B) do lago do Rincón del Bonete. Foram afetados ovinos de todas as idades, exceto cordeiros lactentes. Os primeiros sinais clínicos ocorreram no início de outubro e as mortes de dezembro a fevereiro. Morbidade de 10% foi observada nos Surtos A e B. A mortalidade foi de 7,2% e 2,8% nos Surtos A e B, respectivamente. Os sinais clínicos incluíram perda de peso, abdômen retraído, marcha rígida e cifose. Foram necropsiados um animal de cada rebanho. Observou-se mineralização arterial e pulmonar, nefrocalcinose e osteopetrose no exame macroscópico e histológico dos dois ovinos. Hiperplasia e carcinoma de células C da tireoide foram observados no ovino A. O ovino B apresentou hiperplasia de células C da tireoide e atrofia das células principais da paratireoide. As paratireoides do ovino A não foram examinadas. O diagnóstico diferencial da calcinose enzoótica no Sul da América do Sul deve considerar quatro plantas calcinogênicas da família Solanaceae: Solanum glaucophyllum, Solanum stuckertii, Nierembergia veitchii e Nierembergia rivularis.


#2 - Bacteriological, cytological and histopathological evaluation of the reproductive tract of slaughtered cows

Abstract in English:

Reproductive diseases, mainly endometritis, are important hurdles in cattle raising, In the current study we evaluated gross, bacteriological, cytological, and histological findings from selected sites of the genital from 23 slaughtered cows and tested whether there is an association between these findings and the probability of reaching a reliable diagnosis. The results from the examinations of macroscopic aspects of uterine secretions, the cytological, bacteriological, and histopathological findings were then correlated. There was no significant correlation (P>0.05) of the statistical data from different parts of the genital tract. Trueperella pyogenes and Escherichia coli were isolated from the vagina in 3/23 cases. In only 2/23 samples Enterococcus faecalis and a gram-negative, oxidase-positive bacteria were isolated from the cervix uteri. Only Staphylococcus epidermidis, in 1/23 case, was isolated from the uterus. Histopathological findings in uterus from samples of Groups II (moderate lesions) and III (severe lesions) did not translated in grossly visible changes. Samples from reproductive tracts with secretion in the vagina and cervix uteri had no detectable changes in the other parameters analyzed from this portion. Uterus with positive bacterial culture had evidence of ascendant inflammation judging by the high granulocyte count in the three analyzed portions. This study reinforces that vaginitis and cervicitis in the cow diagnosed only by clinical examination does not reflect the real status of the uterine health. For this reason, treatment of uterine disorders should be conducted based on reliable tests to determine the appropriate therapy for each situation.

Doenças reprodutivas causam altas perdas econômicas nos rebanhos bovinos, sendo a endometrite uma das alterações mais relevantes. Os objetivos desta pesquisa foram avaliar a associação dos achados fisiopatológicos em exames macroscópicos, bacteriológicos, citológicos e histopatológicos nas porções do trato genital de 23 vacas abatidas, bem como avaliar a necessidade de associação destes exames para efetuar diagnóstico fidedigno. A avaliação macroscópica da secreção, os exames histopatológico, citológico e as bactérias identificadas foram correlacionados. Não houve associação (P>0,05) dos resultados nas diferentes porções do trato genital. Na vagina foram isoladas as bactérias Trueperella pyogenes e Escherichia coli. Na cérvix, em apenas 2/23 (8,6%) amostras isolou-se Enterococcus faecalis e gram negativo oxidase positiva. No útero houve isolamento apenas da bactéria Staphylococcus epidermidis. As amostras histopatológicas classificadas em grupo II e III não apresentaram alterações detectadas no exame macroscópico. As amostras com secreção não fisiológica na vagina e cérvix não apresentaram alterações nos outros exames na porção uterina. A amostra com cultura bacteriológica positiva no útero demonstrou uma infecção ascendente através da alta contagem de granulócitos nas três porções analisadas. O presente estudo reforça o conceito de que a vaginite e cervicite diagnosticadas pelo exame clinico na vaca não retratam o real status da saúde uterina e por esta razão, o tratamento do útero deve ser conduzido com critério, alicerçado nos exames complementares para definir a terapia adequada para cada situação.

E-mail: rgumes@hotmail.com The epidemiological, clinic and morphological (pathological and ultrastructural) aspects of four outbreaks of copper deficiency affecting 21- to 90-day-old pigs in the Northeast region of Brazil are reported. Clinical signs began with paraparesis and ataxia and progressed to flaccid or spastic paralysis of the pelvic and thoracic limbs, followed by sternal and/or lateral recumbence. In addition, some animals showed dog-sitting position and intention tremors. The clinical manifestation period was 5-20 days. Significant gross lesions were not observed; however, microscopically, symmetrical degeneration of the white matter with ballooned myelin sheaths containing occasional macrophages was observed, mainly in the spinal cord. Two pigs presented with necrosis ad loss of Purkinje cells and ectopic Purkinje cells in the granular layer and cerebellar white matter. A ultrastructural analysis showed different degrees of damage of myelinated axons in the spinal segments, including an absence of the axoplasm structures with only axonal residues remaining. The myelin sheaths were degenerated and often collapsed into the space previously occupied by the axon. These results suggest that myelin degeneration is secondary to the axonal lesion. Finally, the concentration of copper in the liver was determined using atomic absorption spectrophotometry and was found to be low (ranging from 2.2 to 10.8 ppm). In conclusion, in the Brazilian semiarid region, Cu deficiency occurs in 21 to 90-day-old pigs that ingested different types of waste in their food.

Além disso, alguns animais apresentaram posição de cão sentado e tremores de intenção. O período de manifestação clínica variou de 5-20 dias. Não foram observadas lesões macroscópicas significativas; no entanto, microscopicamente, foi observada degeneração simétrica da substância branca com fragmentação das bainhas de mielina, contendo ocasionais macrófagos, principalmente na medula espinal. Dois suínos apresentaram necrose e perda de células de Purkinje e células de Purkinje ectópicos na camada granular da substância branca cerebelar. A análise ultraestrutural mostrou diferentes graus de lesões em axônios mielinizados em segmentos da medula espinhal, incluindo o desaparecimento de estruturas do axoplasma, restando apenas restos axonais. A bainha de mielina encontrava-se degenerada e muitas vezes, colapsada dentro do espaço previamente ocupado pelo axônio. Esses resultados sugerem que a degeneração da mielina é secundária à lesão axonal. Finalmente, a concentração do cobre no fígado foi determinada usando espectrometria de absorção atômica e revelou baixos valores (variando de 2,2-10,8ppm). Conclui-se que na região semiárida do Brasil ocorre deficiência de cobre em suínos de 21 a 90 dias de idade alimentados com diferentes tipos de resíduos.


#4 - Ultrastructural lesions in the myocardium and kidneys of rabbits in experimental acute Amorimia exotropica poisoning, 36(3):161-166

E-mail: gmpsoares@gmail.com Amorimia exotropica is an important plant associated with sudden death in cattle in Southern Brazil. In order to understand the mechanisms by which A. exotropica causes acute lesions in the heart and kidney of intoxicated animals, an experiment was conducted to determine the histopathology and ultrastructure of myocardial and renal lesions of intoxicated rabbits. After receiving 18g/kg of dried plant, six rabbits died suddenly. At necropsy, the liver was swollen and no other macroscopic lesions were observed. Histologically, centrolobular and midzonal hepatocytes were vacuolated. These vacuoles were strong PAS stained positive, suggesting that they corresponded to glycogen accumulations. In some regions of the ventricular septum and ventricles were found vacuoles of different sizes and the kidneys of two rabbits showed vacuolar degeneration on distal convoluted tubules. Ultrastructurally, the myocardium had cardiomyocytes swelling with separation of myofibrils bundles and rupture and disorganization of the sarcomeres. The mitochondria displayed swelling, disorganization, disruption of the mitochondrial cristae, and electron-dense matrix. Some mitochondria exhibited eccentric projections of their membranes with disruption of both outer and inner membranes. The sarcoplasmic reticulum had no alterations, whereas the T-tubule system was occasionally dilated and ruptured. The kidneys had mitochondrial swelling with disorganization and disruption of the mitochondrial cristae. The vacuoles result from the swelling of the endoplasmatic reticulum and usually were located between two basolateral infoldings and mitochondria, occurring preferentially around the nucleus. The myocytes and T system damages induced by A. exotropica result in acute heart failure and death. Furthermore, this mechanism of cardiotoxicity may be common to all plant containing monofluoroacetate.

Visando compreender os mecanismos pelos quais a A. exotropica provoca lesões agudas no coração e rins de animais intoxicados, foi conduzido uma intoxicação experimental em coelhos para determinar a histopatologia e ultraestrutura da lesão miocárdica e renal. Depois de receber 18g/kg de planta seca, seis coelhos morreram subitamente. Na necropsia, o fígado apresentava acentuação do padrão lobular. Os demais órgãos não apresentaram alterações macroscópicas. Histologicamente, os hepatócitos centrolobulares e mediozonais estavam vacuolizados e coraram-se fortemente com PAS. Em algumas regiões foram observados vacúolos de diferentes tamanhos no septo ventricular e ventrículos e os rins de dois coelhos mostraram degeneração vacuolar nos túbulos contorcidos distais. Ultraestruturalmente, o miocárdio apresentou cardiomiócitos tumefeitos com separação das bandas de miofibrilas e ruptura e desorganização dos sarcômeros. As mitocôndrias estavam tumefeitas exibindo desorganização das cristas mitocondriais, e a matriz estava eletrodensa. Algumas mitocôndrias exibiam projecções excêntricas das suas membranas com ruptura das membranas externas e internas. O retículo sarcoplasmático não tinha alterações, e os túbulos T estavam ocasionalmente dilatados e rompidos. Os rins apresentavam tumefação mitocondrial com desorganização e ruptura das cristas mitocondriais. Os vacúolos resultam da expansão do retículo endoplasmático e foram localizados geralmente entre duas invaginações basolaterais e as mitocôndrias, ocorrendo preferencialmente ao redor do núcleo. A lesão nos miócitos e o dano no sistema T induzido pela A. exotropica resultam na insuficiência cardíaca aguda e morte. Este mecanismo de cardiotoxicidade pode ser comum a todas as plantas contendo monofluoroacetato.

The vacuolization can be caused by intramyelinic edema, as in maple syrup urine disease and some toxic diseases, or by astrocytic edema, as in citrullinemia. This paper reports a spongy degeneration in two Sindhi calves, a female and a male. Both calves were offspring from the same bull, and the herd had high degree of inbreeding. The female calf was born normal, but progressive nervous signs were observed since 2 months old. The male calf had progressive nervous signs since it was born. Both calves were euthanized with severe nervous signs when 4 months old. At necropsy the liver was pale in the male calf. Histological alterations were mild to accentuated vacuolization, mainly in the deep cerebral cortex, cerebellar white matter, grey matter of the spinal cord, internal capsule, and brain stem. Fatty degeneration was observed in the liver of both animals. On electron microscopy it was found that the status spongiosus was caused by astrocytic edema. It is suggested that the disease is caused by a metabolic hereditary error.

All affected calves were sired by the same bull. Necropsy was performed on three affected calves. The only gross lesion detected was paleness of the skeletal muscles of the trunk and limbs. Cytoplasmic vacuoles, the main histological lesion, were particularly evident in skeletal muscles, myocardium and Purkinje fibers, in neurons of the brain and spinal cord. Large amounts of periodic acid Schiff (PAS) positive granules were also observed in these most severely affected tissues. Pretreatment with diastase completely abolished the PAS reactivity. The 1057?TA, a lethal mutation in the gene of the acid alpha-glucosidase, which causes generalized glycogenosis in Brahman cattle, was detected by PCR in paraffin embedded tissues of affected animals on which post-mortem examination was performed. Clinical, histological and molecular findings were similar to previous descriptions of generalized glycogenosis in Brahman cattle in Australia. No previous indexed reports about generalized glycogenosis of Brahman cattle in Brazil could be found.

#7 - Intoxicação experimental por Tetrapterys multiglandulosa (Malpighiaceae) em ovinos, p.91-96

E-mail: griet@ufpa.br Cardiac fibrosis was observed in a calf showing dullness, weakening and respiratory insufficiency in a farm in the state of São Paulo, where cardiac insufficiency, abortion and nervous signs in cattle were associated with the ingestion of Tetrapterys multiglandulosa. The objectives of this paper were to determine the susceptibility of sheep to the intoxication by T. multiglandulosa, to describe the clinical and pathological characteristics of the intoxication, and to evaluate the possibility of using sheep as an experimental species for toxicological studies with this plant. In a previous experiment to determine the toxicity of T. multiglandulosa to be used in sheep, the green plant was given to a steer at the dose of 22g per kg body weight (g/kg/bw), daily, during 9 days. After 9 days the steer showed nervous signs, and on the 12thday was euthanatized. No gross lesions were observed at necropsy. Status spongiosus was observed on the deeper layers of the cerebral cortex and subcortical white matter. Six male sheep were divided in three groups of two sheep each. Group 1 (Sheep 1 and 2) received daily doses of 6 g/kg/bw of the dry plant, during 30 days; Group 2 (Sheep 3 and 4) received daily doses of 3 g/kg/bw, during 60 days; and Group 3 was the control group. Sheep 1 was euthanatized 30 days after the start of the ingestion. Only cardiac arrhythmia was observed clinically, and no lesions were observed at necropsy. Sheep 2, 3 and 4 had also cardiac arrhythmia from day 9, 12 and 18, respectively. From day 52 they started to show depression, reluctance to move and incoordination. Clinical signs got gradually worst and the sheep were euthanatized on days 60, 70 and 80, when clinical signs were marked, and the animals will die at any moment. Hydrothorax, hydropericardium, ascites, nutmeg appearance of the liver and hard whitish myocardium, mainly in the interventricular septum and left ventricle, were observed at necropsy. Histologically, the heart of Sheep 2, 3 and 4 had areas of fibrosis associated with mononuclear cell infiltration. No lesions were found in the heart of Sheep 1. The 4 treated sheep had status spongiosus in different areas of the cerebrum and brain stem, mainly of the deep layers of the cerebral cortex and subcortical white matter. Status spongiosus were also observed in the cerebellar white matter and cervical medulla. In the later the vacuolization was moderate in the white matter and mild in the grey matter. E-mail: griet@ufpa.br Cardiac fibrosis was observed in a calf showing dullness, weakening and respiratory insufficiency in a farm in the state of São Paulo, where cardiac insufficiency, abortion and nervous signs in cattle were associated with the ingestion of Tetrapterys multiglandulosa. The objectives of this paper were to determine the susceptibility of sheep to the intoxication by T. multiglandulosa, to describe the clinical and pathological characteristics of the intoxication, and to evaluate the possibility of using sheep as an experimental species for toxicological studies with this plant. In a previous experiment to determine the toxicity of T. multiglandulosa to be used in sheep, the green plant was given to a steer at the dose of 22g per kg body weight (g/kg/bw), daily, during 9 days. After 9 days the steer showed nervous signs, and on the 12thday was euthanatized. No gross lesions were observed at necropsy. Status spongiosus was observed on the deeper layers of the cerebral cortex and subcortical white matter. Six male sheep were divided in three groups of two sheep each. Group 1 (Sheep 1 and 2) received daily doses of 6 g/kg/bw of the dry plant, during 30 days; Group 2 (Sheep 3 and 4) received daily doses of 3 g/kg/bw, during 60 days; and Group 3 was the control group. Sheep 1 was euthanatized 30 days after the start of the ingestion. Only cardiac arrhythmia was observed clinically, and no lesions were observed at necropsy. Sheep 2, 3 and 4 had also cardiac arrhythmia from day 9, 12 and 18, respectively. From day 52 they started to show depression, reluctance to move and incoordination. Clinical signs got gradually worst and the sheep were euthanatized on days 60, 70 and 80, when clinical signs were marked, and the animals will die at any moment. Hydrothorax, hydropericardium, ascites, nutmeg appearance of the liver and hard whitish myocardium, mainly in the interventricular septum and left ventricle, were observed at necropsy. Histologically, the heart of Sheep 2, 3 and 4 had areas of fibrosis associated with mononuclear cell infiltration. No lesions were found in the heart of Sheep 1. The 4 treated sheep had status spongiosus in different areas of the cerebrum and brain stem, mainly of the deep layers of the cerebral cortex and subcortical white matter. Status spongiosus were also observed in the cerebellar white matter and cervical medulla. In the later the vacuolization was moderate in the white matter and mild in the grey matter. The minimal amount of the dry plant consumed was 11 g/kg and the maximum was 30 g/kg. The progression of clinical findings was similar in six animals with slight diarrhea at 20 days of experiment. Neurological signs were observed at 25 days and included ataxia with dysmetria, muscle tremors of the head, atypical postural reactions, frequent falls, sluggish of movements, difficulty in grazing and swallowing. These signs were enhanced when the animals were forced to walk. Four of the animals presented progressive emaciation. The sheep whose supply of the plant was interrupted recovered gradually, and 11 days after the animal returned to normal. During necropsy, only enlarged mesenteric lymph nodes were observed. The histological alterations were more significant in the central nervous system, with multiple and severe cytoplasmic distention and vacuolation which affects specially Purkinje cells of the cerebellum, neurons of cerebral cortex, thalamus, midbrain and the ventral horn of spinal cord. Three cows in the 8th month of pregnancy received daily doses of 1-2 g/kg of the leaves, until a total amount of 10, 21 and 28 g/kg/bw was reached. Two 8-month-pregnant cows were fed 15.5 and 18 g/kg of the dried leaves. Two 4-month-pregnant cows were fed 35 g/kg of green leaves of A. glazioviana. The cow treated with 21 g/kg of the dry plant material showed clinical signs of poisoning and delivered a stillborn calf. No clinical signs were observed in the other cows. The calves from cows that received 9, 15.5 and 28 g/kg of the dried leaves showed weakness and suckling difficulties, and were killed. Whitish areas and thickening of the right ventricle wall of the heart were observed in the stillborn calf. Thickening of the wall of the right ventricle was also observed in the calf delivered by the cow treated with 28g/kg of dry plant. The other calves had no gross lesions. The histological changes in all necropsied calves were characterized by tumefaction and vacuolization of muscle fibers and proliferation of fibrous tissue, similar as occurred in spontaneous cases of fibrosis of the heart muscle in cattle poisoned by A. glazioviana. E-mail: tokarnia@ufrrj.br Due to doubts which still persist 25 years after the outbreak of African swine fever (ASF) which ocurred in the county of Paracambi, Rio de Janeiro, Brazil, in 1978, the results obtained through the studies to establish and confirm the diagnosis are presented. These include data on the epidemiology, clinic-pathological aspects, bacteriological, virological and ultramicroscopic examinations, the experimental reproduction of the disease and cross immunity tests with classical swine fever virus performed in Brazil, and on the confirmation with isolation of the virus and determination of its identity at the Plum Island Animal Disease Center, New York, USA. The pigs of the affected herd had been fed untreated remains of meals from airplanes of international lines flying to Brazil from Portugal and Spain where ASF was occurring at the time. According to publication by the Ministry of Agriculture, after the diagnosis of the outbreak of ASF described in this paper, 223 additional outbreaks were reported in Brazil between 1978 and 1979, in all the Brazilian regions (North, Northeast, Central-West, Southeast and South). Further outbreaks were reported in 1981, but their number is not known. The last case was reported to have occurred on November 15, 1981, and on December 5, 1984, Brazil was declared free of ASF. For the diagnosis of ASF 54,002 samples were examined by the Department of Virology of the Institute of Microbiology, Federal University of Rio de Janeiro, from 1978 to 1981, by the techniques of haemadsorption in leucocyte cultures (HAd), direct immunoflorescence in tissue sections (FATS), direct immunoflorescence in cell cultures (FATCC), immuno-electro-osmophoresis (IEOP) and indirect immunoflorescence assay (IIF). Only 4 samples were positive with the FATCC procedure. This is the only technique which includes virus isolation; the origin of these positive samples was not reported, but probably they were from the Paracambi outbreak. From other suspected outbreaks of ASF in Brazil there is no information on the isolation and characterization of the virus isolates. Likewise there is no information available about the epidemiology, clinical signs, and pathology of suspected ASF in other outbreaks.
